Fundada em 1831, a Biblioteca Central da Universidade de Humboldt é uma das mais antigas bibliotecas de Berlim. Desde cedo foi-lhe concedido o direito de depósito legal relativo a publicações de Brandengurg e Berlim, tendo este direito cessado em 1972.
Uma vez que as colecções não foram muito danificadas durante a Segunda Guerra Mundial, o fundo da Biblioteca é constituído maioritariamente por livros que datam de 1850 a 1945. Com o objectivo de expandir a colecção de livros antigos, foram adquiridas bibliotecas eruditas especializadas, como por exemplo os 6.000 volumes da biblioteca privada de Jacob e Wilhelm Grimm, parte da biblioteca filosófica de Wilhelm von Humboldt (250 volumes) e a colecção Theodor Schmalz.
Outros tesouros da Biblioteca Central são constituídos por partes da ex-biblioteca do Príciple Oskar da Prússia, e por um vasto conjunto de cadernos de apontamentos de aulas (cerca de 50.000 títulos).
As bibliotecas departamentais também possuem importantes colecções, como a antiga biblioteca da 'Gesellschaft der Naturforschenden Freunde zu Berlin', fundada em 1773 (aproximadamente 1.000 livros) e a biblioteca privada de August Wilhelm von Hofmann (cerca de 8.000 livros), o fundador do primeiro instituto químico de Berlim. As bibliotecas de Humanidades possuem importantes colecções provenientes do Departamento de História de Arte.

There are a total of approximately 6 million volumes in the holdings of the Library.
